CervoMed presents new neflamapimod data in DLB at AAIC 2026

CervoMed announced new clinical, plasma biomarker, and imaging data for neflamapimod in dementia with Lewy bodies at the Alzheimer's Association International Conference 2026. New analyses from the 159-patient Phase 2b RewinD-LB trial indicate that the failure to replicate earlier Phase 2a results was due to a higher-than-targeted proportion of patients with Alzheimer's disease co-pathology and use of a drug batch that did not achieve expected plasma concentrations. Exploratory analyses showed neflamapimod slowed worsening of DLB in patients with low plasma pTau181 and in those who achieved expected drug levels, with a consistently improving treatment effect at progressively lower pTau181 levels. Neflamapimod also demonstrated durable slowing of right basal forebrain atrophy and increased basal forebrain connectivity relative to placebo over 48 weeks. A separate Phase 2 study of an 80 mg twice-daily dose met its primary safety, tolerability, and pharmacokinetic objectives, and a pharmacokinetic-pharmacodynamic analysis identified a plasma trough threshold of about 4 ng/mL associated with clinical improvements, supporting selection of a 50 mg three-times-daily dose for the planned Phase 3 trial.

CervoMed Completes Enrollment in Phase 2 Study of Neflamapimod for Nonfluent Variant Primary Progressive Aphasia

CervoMed has completed enrollment in its Phase 2a study of neflamapimod for nonfluent variant primary progressive aphasia, a type of frontotemporal dementia. The trial enrolled 25 participants who will receive oral neflamapimod for 24 weeks, followed by a 12-week randomized, double-blind placebo-controlled extension. Interim biomarker data from the study will be presented at the 19th Clinical Trials on Alzheimer's Disease conference in Boston in November 2026, with first clinical data expected in the first quarter of 2027.
